- Characterization of lipid droplets in Malassezia spp: unraveling the role in the lipid metabolism

Interest in the study of lipid droplets in other organisms than humans is increasing. Our previous results revealed that LDs are present and can be detected in Malassezia spp. However, LD structure and dynamics seem very different from LDs detected in human cells, even their morphology is very different. The lipid dependence in Malassezia genus makes this organelle very attractive to study and to characterize. We aim to understand the physiological relevance of LDs and the dynamic behavior of LDs in lipid homeostasis and metabolism. In addition, their contribution to cellular membrane and organelle integrity, stress response, prevention of lipotoxicity, growth and survival will be investigated. The main goal of this proposal is to characterize LDs from Malassezia spp by in silico genomic analysis, in combination with lipidomics and proteomics. Additionally, we will evaluate LD morphology and dynamics using live-cell imaging (confocal microscopy) and detailing the ultrastructure with transmission electron microscopy (TEM). The detection of the surface proteins in LDs will be performed by tagging perilipin protein using Agrobacterium-mediated genetic transformation system by a binary T-DNA vector with the green fluorescent protein gene (gfp). This research will allow us to investigate LD from Malassezia spp. at the cellular and molecular level, which may become highly relevant to understand and make assessments to the physiological regulation of global lipid metabolism in this yeast. Additionally, lipidomic and proteomic data integration will be carried out in order to contextualize lipid metabolism based on LD using a previously reported metabolic model. This contextualization will allow us to understand the underpinnings behind metabolic flow distribution in the dynamic of LD lipid content to have a better understanding of the regulatory systems, underlying lipid functions and interactions in the context of cellular and physiology aspects in the yeast.
